本シンポジウムはアジア太平洋地域におけるプラスチック汚染の危機に対するプラスチックのリサイクル/アップサイクルを実現する革新的な研究をテーマとしたシンポジウムになります。
津田は”Industrial Enzyme Discovery and Engineering from Microbial Single-cell Genome Database”と題して、当社の独自の微生物遺伝子データベースbit-GEMと計算科学を用いた酵素開発プラットフォームbit-QEDによる、プラスチック分解酵素の開発について話します。
＜Abstract＞
Environmental microbiota represent a vast, untapped resource for novel biocatalytic enzymes. We have developed a proprietary microbial genome database using a microbial single-cell sequencing method named bit-MAP(R). This method efficiently recovers microbial genes from diverse environmental microbiota (including soil, hot springs, and seawater) that are difficult to obtain with existing sequencing technologies.
To capitalize on this resource, we developed a computational pipeline, bit-QED, to identify enzymes capable of specific reactions by analyzing the active sites of enzymes. We screened the bit-GEM database using the proprietary method, and identified a number of unique PET-degrading enzymes that are phylogenetically distinct from existing ones. Furthermore, we optimized a PET-degrading enzyme via in silico modeling-guided directed evolution. Within a few rounds of protein engineering, our enzymes were able to degrade PET films at ambient temperatures, suggesting their potential for efficient and eco-friendly plastic recycling. The discovery of numerous novel PET-degrading enzymes demonstrates the utility of our bit-GEM database. Leveraging bitBiome’s extensive microbial genetic information, we aim to unlock new possibilities for industrial enzymes from unknown microorganisms.
